Our comprehension of the phenotypical and functional heterogeneity of human all-natural killer (NK) cells has significantly improved over the previous couple of decades. This progress has been greatly aided by the growth of culture conditions such as clonal growth of NK cells. Analysis of individual NK cell clones has caused the original observation that distinct NK cell clones recognize different significant histocompatibility complex (MHC) class I specificities. Engagement of Ly49 receptors using their individual MHC ligands leads to negative regulation of NK cell effector functions, consistent with a crucial function of the receptors in”itself” recognition. The finding that many NK cells which express the Ly49A receptor do this from one Ly49A allele (whereby expression may happen from the maternal or the paternal chromosome) may thus reflect a putative receptor supply process that limits the Amount of Ly49 receptors expressed in One NK cell
